Studies indicate that approximately 18-30% of cell lines are cross-contaminated or misidentified. The use of such compromised cells can lead to erroneous research conclusions, irreproducible results, and potentially catastrophic outcomes in clinical cell therapy. Since 2009, the NIH and ATCC have urged researchers to perform STR profiling and have initiated the development of cell line STR databases. Today, STR profiling is the gold standard for authenticating cell line identity and detecting cross-contamination. Most authoritative journals now require authors to provide STR authentication data upon manuscript submission. The Chinese Pharmacopoeia explicitly mandates identity testing for newly established cell lines/strains, cell banks, and end-of-production cells to confirm their unique identity and absence of cross-contamination.
